Prelude CEO Highlights Strong Execution in First Half of 2026
"The first six months of 2026 were highlighted by steady and strong execution across our organization," stated Kris Vaddi, Chief Executive Officer of Prelude. "We've made considerable progress advancing our three core programs. Notably, we are well positioned to initiate, in the fourth quarter, the first clinical trial of our highly differentiated, selective KAT6A degrader, PRT13722 in HR+ breast cancer. Enrollment in our phase 1 study of PRT12396, our mutant-selective JAK2V617F inhibitor, continues, and we are also making excellent progress toward advancing the lead development candidates from our mCALR degrader antibody conjugate program."
Prelude Therapeutics Shares Rise 20% to $4.67
Shares of Prelude Therapeutics (PRLD) after moving higher after Incyte (INCY) in its Q2 earnings release that after a review of available data, it has discontinued further development of INCB160058 to prioritize its next-generation JAK2V617F-targeted pipeline. On November 4, 2025, Prelude announced an exclusive option agreement with Incyte focused on Prelude's mutant selective JAK2V617F JH2 inhibitor program in development for patients with myeloproliferative neoplasms. Per the agreement, Incyte has an exclusive option to acquire the JAK2V617F program in exchange for an upfront payment and a strategic equity investment in Prelude, plus potential downstream milestones and royalties. Shares of Prelude Therapeutics are up 20% to $4.67 in morning trading. Incyte is up 6% to $125.48 following its earnings report.
Prelude CEO States Focused Execution of Strategic Priorities
"Through this first quarter of 2026, our company has continued to demonstrate focused execution of the strategic priorities we set forth late last year." stated Kris Vaddi, Chief Executive Officer of Prelude. "Since the beginning of this year, we've advanced PRT12396 into first-in-human studies, presented promising preclinical data from our highly selective KAT6A degrader development candidate, continued to progress towards a development candidate from our mCALR program and importantly, extended our cash runway into the second quarter of 2028."
Prelude Therapeutics Presents New Data on PRT13722
Prelude Therapeutics announced the presentation of new preclinical data from its lead development candidate, PRT13722. PRT13722 is being developed for the treatment of hormone receptor positive, or HR+,/human epidermal growth factor receptor 2, or HER2-, breast cancer, or BC. Based on preclinical data, we believe PRT13722 is a highly differentiated, first-in-class, orally bioavailable, potent and highly-selective KAT6A degrader. Details on the poster presentation are as follows: Title: First-in-Class potent and selective oral KAT6A degrader development candidate, PRT13722, drives complete tumor regressions as a monotherapy with an improved preclinical hematological safety profile. Abstract Control Number: 7335. Session Title: Proximity-Induced Drug Discovery 2. Session Start Time: 4/21/2026 2:00 PM PT Location: Poster Section 15. Summary: PRT13722 is a highly differentiated, first-in-class, orally bioavailable, potent and highly selective KAT6A degrader development candidate. PRT13722, by degrading KAT6A, drives more complete disruption of KAT6A regulatory pathways than dual KAT6A/B inhibitors, resulting in more robust depth and breadth of preclinical efficacy in HR+/HER2- breast cancer. KAT6 is an emerging and recently validated target in the treatment of ER+ breast cancer. Prelude discovered and is developing first-in-class, highly potent, highly selective and orally bioavailable KAT6A selective degraders. PRT13722 remains on track for an IND filing in mid-2026 and subject to clearance, with Phase 1 study initiation planned in the 2nd half of 2026. Prelude believes that selectively degrading KAT6A has the potential for improved efficacy, tolerability and combinability with other agents relative to non-selective inhibitors of KAT6A/B.
