Alector Enters Exclusive Licensing Agreement with Genentech
Alector (ALEC) announced it has entered into an exclusive global licensing agreement with Genentech, a member of Roche (RHHBY), granting Genentech worldwide rights to develop and commercialize Alector's AL050 program, its brain-penetrant engineered glucocerebrosidase ERT. AL050 is designed to address GCase deficiency, a driver of neurodegeneration in Parkinson's Disease that is most pronounced in - but not limited to - carriers of GBA1 mutations. It combines an Alector-engineered GCase, optimized for greater enzymatic activity and a longer half-life, with Alector's ABC, which is designed to transport therapeutics across the blood-brain barrier. Delivered into the brain, the enzyme is intended to break down glucosylsphingosine and glucosylceramide, lipids that accumulate in neurons and other brain cells upon GCase deficiency and contribute to neurodegeneration, with the aim of reducing cellular dysfunction and slowing disease progression. Under the terms of the agreement, Alector will receive an upfront payment of $100M and is also eligible to receive additional payments of up to $1.17B upon the achievement of development, regulatory, and commercial milestones. Alector is also eligible for tiered royalties on net sales. Under the collaboration, Genentech will be responsible for development, regulatory, manufacturing, and commercialization across all indications. Alector retains ownership of ABC and full rights to apply the platform across its wholly owned pipeline.
Alector Reports Q2 Revenue of $3.32M, Beating Consensus
Reports Q2 revenue $3.32M, consensus $1.71M. "We continue to make steady progress across our ABC-enabled preclinical and research pipeline," said Arnon Rosenthal, Chief Executive Officer of Alector. "Our work across antibodies, enzymes, and siRNA reflects the versatility of the ABC platform and our strategy to apply it to programs where enhanced brain delivery may address important limitations in the treatment of neurodegenerative diseases. We remain focused on disciplined execution as we advance our portfolio toward clinical development, with the goal of addressing important unmet needs for patients living with neurodegenerative diseases."
Alector Reports Q1 Revenue of $1.05M, Below Consensus of $11.03M
Reports Q1 revenue $1.05M, consensus $11.03M. "Over the past seven years, we have built a highly differentiated blood-brain barrier platform with the versatility to deliver antibodies, enzymes, proteins, and siRNA to the brain," said Arnon Rosenthal, Chief Executive Officer of Alector. "We have a rich and growing pipeline of ABC-enabled programs, and we remain deeply committed to developing therapies that can make a meaningful difference for patients living with neurodegenerative diseases."
Alector Discontinues Phase 2 Trial of Nivisnebart for Early Alzheimer's Disease
Alector (ALEC) confirmed that the Phase 2 PROGRESS-AD trial of nivisnebart in individuals with early Alzheimer's disease will be discontinued. The decision follows a pre-specified futility analysis conducted by an IDMC, which concluded that the trial was unlikely to meet its primary endpoint of slowing disease progression at completion. "This outcome is disappointing for patients and families affected by Alzheimer's disease and underscores the complexity of developing effective treatments for this devastating disease," said Arnon Rosenthal, CEO of Alector. "We are deeply grateful to the patients, caregivers, investigators and study staff who participated in the PROGRESS-AD trial and contributed to advancing the scientific understanding of progranulin biology in neurodegeneration. We remain committed to progressing our broader pipeline of programs targeting neurodegenerative disease, including multiple wholly owned candidates enabled by our ABC platform." Alector and GSK (GSK) have been co-developing nivisnebart. The companies will work closely with study investigators to inform participants enrolled in the PROGRESS-AD trial. Full results will be shared at a future medical meeting or congress.
