Iovance Updates IOV-LUN-202 Trial Results, 24.6% Patient Response
Iovance Biotherapeutics announced updated results from Cohorts 1 and 2 of its Phase 2 IOV-LUN-202 trial and a biomarker-enriched precision tumor infiltrating lymphocyte, or TIL, strategy for lifileucel in previously treated non-squamous metastatic non-small-cell lung cancer, or mNSCLC. As of the October 14 data cutoff, 17 of 69 patients responded, representing an objective response rate, or ORR, of 24.6%, consistent with the 25.6% ORR previously reported in 39 patients. The ORR was 43.3% in the Kirsten rat sarcoma viral oncogene homolog, or KRAS, wild-type subgroup, compared with 11.4% in the KRAS-mutant subgroup. A biomarker-enriched subgroup defined by KRAS wild-type status, human epidermal growth factor receptor 2 wild-type status, and current or former smoking accounted for 13 of 17 observed responses, with an ORR of 59.1%. IOV-LUN-202 Cohorts 1 and 2 are completing enrollment and support the planned supplemental Biologics License Application for lifileucel in non-squamous mNSCLC in 2027. Iovance will prospectively evaluate this novel approach with POSITILITY, a biomarker-enriched cohort designed with registrational intent, with enrollment to begin in November. "These findings are reshaping how we develop TIL therapy for mNSCLC. The expanded dataset reinforces the activity of lifileucel in previously treated disease, while the concentration of responses in KRAS wild-type tumors points to a more precision approach to patient selection. In the enriched subgroup, nearly six in ten patients responded, including two complete responses. We believe these insights could expand lifileucel's potential as a one-time treatment for mNSCLC, with a best-in-class profile compared to other therapies in development," said Frederick Vogt, interim president and CEO of Iovance.
Iovance's Lifileucel Monotherapy Shows Unprecedented Efficacy in Soft Tissue Sarcomas
Iovance announced that one-time lifileucel monotherapy demonstrated unprecedented efficacy with deep, durable responses across two difficult-to-treat soft tissue sarcomas with limited treatment options, metastatic undifferentiated pleomorphic sarcoma and dedifferentiated liposarcoma, in a pilot trial presented orally at the European Society for Medical Oncology Congress 2026 in Madrid. Among the first eight evaluable patients, physician-assessed confirmed objective response rate was 50%. Four partial responses were all ongoing as of the data cut-off on September 9. The safety profile was consistent with the lifileucel treatment regimen across other indications. "On the heels of this pilot trial, we are rapidly enrolling the SARATOGA registrational study to support two new approvals for lifileucel," said Brian Gastman, executive VP, translational medicine and research.
