Roivant and Priovant Announce Phase 3 Results for Brepocitinib
Roivant and Priovant Therapeutics announced results from the Phase 3 CLARITY program evaluating brepocitinib in non-infectious uveitis, or NIU. Brepocitinib met the primary endpoint for both the CLARITY-1 and CLARITY-2 studies. Brepocitinib also achieved statistically significant and clinically meaningful benefit on all key secondary endpoints, including measurements of sustained steroid-free quiescence at Week 48, chorioretinal vascular leakage, resolution of macular edema and patient-reported visual functioning. These results were achieved despite a substantially more aggressive steroid taper than precedent NIU trials. Brepocitinib demonstrated consistent benefits on preventing treatment failure across key subgroups, including intermediate, posterior, and panuveitis; patients with and without prior TNFi experience; and multiple etiologies of uveitis, including idiopathic disease, birdshot chorioretinopathy, and sarcoidosis. Pre-specified multiplicity controlled secondary endpoints in the CLARITY trials included sustained steroid-free quiescence, improvement in chorioretinal vascular leakage as assessed through wide-field fluorescein angiography, resolution of macular edema, and improvement in patient-reported visual functioning. Brepocitinib was generally well tolerated during the treatment period. Most events were mild or moderate in severity, and only 3% of participants discontinued treatment due to an AE. The observed safety profile in the trials was consistent with the known safety profile of brepocitinib, with no new safety signals observed. Priovant intends to file a supplemental New Drug Application, or sNDA, for brepocitinib for treatment of NIU in the first half of calendar year 2027.
H.C. Wainwright: Roivant IMVT-1402 Development Fails
H.C. Wainwright analyst Douglas Tsao notes that this morning, Roivant Sciences (ROIV) reported that IMVT-1402 missed the Week 12 Cutaneous Lupus Erythematosus Disease Area and Severity Index-Activity primary endpoint in a 57-patient cutaneous lupus erythematosus proof-of-concept study and will discontinue development in CLE. While the firm acknowledges this is disappointing, it views it as an indication-specific failure in a biologically complex skin-dominant autoimmune disease. Wainwright remains confident in '1402 in more direct pathogenic-IgG biology, including myasthenia gravis, chronic inflammatory demyelinating polyneuropathy and Graves' disease. The company reiterated to Wainwright that Immunovant (IMVT) - which is majority-owned subsidiary of Roivant - applies a high clinical and competitive bar when deciding which indications to advance, reflecting its focus on capital efficiency. The firm has a Buy rating on Immunovant and a price target of $41 on the shares.
Roivant Halts Immunovant CLE Development
Roivant announced topline results from Period 1 of Immunovant's proof-of-concept trial evaluating IMVT-1402 for the treatment of cutaneous lupus erythematosus, CLE. The study did not achieve statistical significance on its primary endpoint of percent change from baseline in the Cutaneous Lupus Erythematosus Disease Area and Severity Index Activity score at Week 12. Numerical trends favoring IMVT-1402 over placebo were observed across multiple endpoints, and patients who achieved deeper IgG reductions from baseline were more likely to achieve improved clinical responses. However, due to the competitive landscape and the clinical results observed, Immunovant plans to stop development in CLE.
