Lipocine Announces Phase 3 Trial Results for LPCN 1154
Lipocine announced topline results from its Phase 3 placebo-controlled trial evaluating LPCN 1154 - oral brexanolone - for the treatment of postpartum depression, or PPD. LPCN 1154 did not show a statistically significant reduction from baseline in HAM-D total score compared to placebo at hour 60 in the full analysis set and the primary was not met. The results showed LPCN 1154 to be well tolerated and demonstrated a favorable safety profile to support outpatient administration without the need for healthcare provider monitoring. No treatment-related severe or serious adverse events were reported; no cases of excessive sedation or loss of consciousness were observed; and no treatment-related discontinuations were reported. The company identified signals that could indicate a potential development path for LPCN 1154. The company has applied for breakthrough therapy and fast track designations for LPCN 1154 in patients with PPD. While the company is waiting for feedback from the FDA on such designations, the company plans to preserve capital and engage with stakeholders, including investors, regulators, and advisors, to evaluate all options available to the company going forward. Such options may include, but are not limited to, continued development of LPCN 1154, including the potential submission of a validation study protocol, development of other product candidates, strategic transactions, partnerships, and other opportunities.
Lipocine Completes Final Patient Visit in Phase 3 Trial of LPCN 1154
Lipocine announced that the last patient has completed the final study visit in its Phase 3 clinical trial evaluating LPCN 1154 for the treatment of postpartum depression. The randomized, double-blind, placebo-controlled study enrolled a total of 90 patients with severe PPD, having an average baseline Hamilton depression rating scale of 28.3. LPCN 1154 has shown a positive safety profile, with all reported nervous system adverse events being mild to moderate in severity. Furthermore, there were no instances of drug discontinuation, excessive sedation, loss of consciousness, or drug-related serious adverse events.

Lipocine announced that enrollment and participant dosing have been completed in its Phase 3 clinical trial evaluating LPCN 1154 for the treatment of postpartum depression. Lipocine Completes Safety Review of LPCN 1154 Clinical Trial
Lipocine announced the completion of a scheduled independent Data Safety Monitoring Board review of its ongoing Phase 3 clinical trial evaluating LPCN 1154 for the rapid relief treatment of PPD. This was the second of two DSMB reviews planned during the study. The DSMB recommended that the trial continue as planned without modification. The DSMB recommendation was based on assessment of available safety data collected from 82 randomized participants, of which 74 had completed dosing. There have been no treatment discontinuations or reports of drug-related serious adverse events. No cases of excessive sedation or loss of consciousness have been reported to date. The one reported case of a dose reduction was due to an adverse event. The study is no longer screening new participants, however the company continues to enroll the additional participants who have met the eligibility criteria. The company is on track to report topline safety and efficacy results early in the second quarter of 2026. "The data generated to date reinforces our confidence in the safety profile of LPCN 1154," said Mahesh Patel, CEO of Lipocine. "We believe LPCN 1154's target profile, including superior tolerability, rapid therapeutic benefit, and a short 48-hour treatment course, has the potential to establish a new and improved treatment paradigm for PPD.
